Electron Tomography Enhanced Electron Microscopy
A solution for high resolution structures in situ 

At the present it is often possible to determine the structure of molecular machines in situ or at atomic resolution, but seldom both together. Resolving the high-resolution structure(s) of molecular machines in their natural context is critical to understand the process of infection by a viral or bacterial pathogen, for example. The central hypothesis of ETEEM is that if supplementary tomographic information were collected after cryoelectron microscopy (cryoEM), it could be leveraged through novel software to assist single-particle alignment and classification to solve a high-resolution structure in situ, even though a structure might not be tractable with single-particle data alone. Sufficient high-resolution information content exists in 2D cryoEM images, but the information is currently inaccessible for single-particle cryoEM analysis because of the surrounding cellular context and other factors. The information can be rendered accessible with the assistance of cryo-electron tomography (cryoET).
Ultimately, we hope can resolve the atomic structure of large macromolecular assemblies in situ, which is to say in an authentic cell or cell-like environment, which may further reveal some fundamental mechanisms underlying virus infection. This method does not restrict type of sample, but does affected by sample thickness that is strongly related to the level of inelastic scattering and the “pink” noise level contributed from cellular context. As the emerging of cryoFIB technique, we expect ETEEM can also be directly applied to many large macromolecular assemblies such as human viruses and intracellular machines involved in cellular function or cancer.
